Slaughter hit a 3-point shot 3:20 into the second half to give Western Kentucky the lead for good at 45-42, but the Hilltoppers led by no more than five until Slaughter made two free throws with 12 seconds left.
The game was part of a double header, the first of which was won by Tennessee over Middle Tennessee 75-54.
Sergio Kerush added 15 points for Western Kentucky (5-3) and Jeremy Evans had 12.
Vanderbilt (6-3) was paced by Jeffery Taylor with 21.
